    Funqual: User-Defined, Statically-Checked Call Graph Constraints in C++

    Static analysis tools can aid programmers by reporting potential programming mistakes prior to the execution of a program. Funqual is a static analysis tool that reads C++17 code ``in the wild\u27\u27 and checks that the function call graph follows a set of rules which can be defined by the user. This sort of analysis can help the programmer to avoid errors such as accidentally calling blocking functions in time-sensitive contexts or accidentally allocating memory in heap-sensitive environments. To accomplish this, we create a type system whereby functions can be given user-defined type qualifiers and where users can define their own restrictions on the call graph based on these type qualifiers. We demonstrate that this tool, when used with hand-crafted rules, can catch certain types of errors which commonly occur in the wild. We claim that this tool can be used in a production setting to catch certain kinds of errors in code before that code is even run

    A murine model of variant late infantile ceroid lipofuscinosis recapitulates behavioral and pathological phenotypes of human disease.

    Neuronal ceroid lipofuscinoses (NCLs; also known collectively as Batten Disease) are a family of autosomal recessive lysosomal storage disorders. Mutations in as many as 13 genes give rise to ∼10 variants of NCL, all with overlapping clinical symptomatology including visual impairment, motor and cognitive dysfunction, seizures, and premature death. Mutations in CLN6 result in both a variant late infantile onset neuronal ceroid lipofuscinosis (vLINCL) as well as an adult-onset form of the disease called Type A Kufs. CLN6 is a non-glycosylated membrane protein of unknown function localized to the endoplasmic reticulum (ER). In this study, we perform a detailed characterization of a naturally occurring Cln6 mutant (Cln6(nclf)) mouse line to validate its utility for translational research. We demonstrate that this Cln6(nclf) mutation leads to deficits in motor coordination, vision, memory, and learning. Pathologically, we demonstrate loss of neurons within specific subregions and lamina of the cortex that correlate to behavioral phenotypes. As in other NCL models, this model displays selective loss of GABAergic interneuron sub-populations in the cortex and the hippocampus with profound, early-onset glial activation. Finally, we demonstrate a novel deficit in memory and learning, including a dramatic reduction in dendritic spine density in the cerebral cortex, which suggests a reduction in synaptic strength following disruption in CLN6. Together, these findings highlight the behavioral and pathological similarities between the Cln6(nclf) mouse model and human NCL patients, validating this model as a reliable format for screening potential therapeutics

    In 1895, Montana adopted a version of the Field Civil Code--a massive law originally drafted by New York lawyer David Dudley Field in the early 1860s. The Civil Code (and its companion Political, Penal, and Procedural Codes) were adopted without debate, without legislative scrutiny, and without Montanans having an opportunity to grasp the enormity of the changes the Codes brought to the Montana legal system. In sponsoring this debate over whether to repeal the Civil Code, the Montana Law Review is finally giving Montana the opportunity to examine the merits of the Civil Code that she was denied 105 years ago

    Do the rat anterior thalamic nuclei contribute to behavioural flexibility?

    The rodent anterior thalamic nuclei (ATN) are vital for spatial memory. A consideration of their extensive frontal connections suggests that these nuclei may also subserve non-spatial functions. The current experiments explored the importance of the ATN for different aspects of behavioural flexibility, including their contribution to tasks typically associated with frontal cortex. In Experiment 1, rats with ATN lesions were tested on a series of response and visual discriminations in an operant box and, subsequently, in a water tank. The tasks included assessments of reversal learning as well switches between each discrimination dimension. Results revealed a mild and transient deficit on the operant task that was not specific to any stage of the procedure. In the water tank, the lesion animals were impaired on the reversal of a spatial discrimination but did not differ from controls on any other measure. Experiment 2 examined the impact of ATN damage on a rodent analogue of the ‘Stroop’, which assesses response choice during stimulus conflict. The lesion animals successfully acquired this task and were able to use contextual information to disambiguate conflicting cue information. However, responding during the initial presentation of conflicting cue information was affected by the lesion. Taken together, these results suggest that the ATN are not required for aspects of behavioural flexibility (discrimination learning, reversals or high-order switches) typically associated with the rat medial prefrontal cortex. The results from Experiment 2 suggest that the non-spatial functions of the ATN may be more aligned with those of the anterior cingulate cortex

    Psychosocial work conditions as determinants of well-being in Jamaican police officers: the mediating role of perceived job stress and job satisfaction

    Policing is considered a high-stress occupation due to the demanding nature of the job. Addressing police stress requires a detailed understanding of how psychosocial risk factors influence various aspects of their well-being. Moreover, an analysis of the direct effects of work conditions and the mediating effects of cognitive appraisals is also warranted. Using the Demands–Resources–Individual Effects (DRIVE) model of work-related stress this study investigated the direct effects of work conditions on well-being and also examined the intermediate role of perceived job stress and job satisfaction in the relationship between work conditions and well-being. Five hundred and seventy-eight police officers from the Jamaica Constabulary Force (J.C.F.) completed the questionnaire. Data were analyzed using hierarchical regressions and the Hayes Process tool for mediation analysis. Both perceived job stress and satisfaction mediated the relationship between work conditions and general physical health. Perceived job stress was an indirect pathway through which work conditions influenced psychological distress, whereas job satisfaction was not a significant mediator. In contrast, job satisfaction mediated the relationship between work conditions and positive well-being, but perceived job stress did not. These findings provide significant evidence for periodically monitoring and auditing perceptions of stress and job satisfaction, as they are likely precursors to subsequent health problems

    A review of Bayesian belief network models as decision-support tools for wetland conservation : are water birds potential umbrella taxa?

    10 pagesCreative approaches to identifying umbrella species hold promise for devising effective surrogates of ecological communities or ecosystems. However, mechanistic niche models that predict range or habitat overlap among species may yet lack development. We reviewed literature on taxon-centered Bayesian belief network (BBN) models to explore a novel approach to identify umbrella taxa identifying taxonomic groups that share the largest proportion of habitat requirements (i.e., states of important habitat variables) with other wetland-dependent taxa. We reviewed and compiled published literature to provide a comprehensive and reproducible account of the current understanding of habitat requirements for freshwater, wetland-dependent taxa using BBNs. We found that wetland birds had the highest degree of shared habitat requirements with other taxa, and consequently may be suitable umbrella taxa in freshwater wetlands. Comparing habitat requirements using a BBN approach to build species distribution models, this review also identified taxa that may not benefit from conservation actions targeted at umbrella taxa by identifying taxa with unique habitat requirements not shared with umbrellas. Using a standard node set that accurately and comprehensively represents the ecosystem in question, BBNs could be designed to improve identification of umbrella taxa. In wetlands, expert knowledge about hydrology, geomorphology and soils could add important information regarding physical landscape characteristics relevant to species. Thus, a systems-oriented framework may improve overarching inferences from BBNs and subsequent utility to conservation planning and management.Postprin

    MicroRNAs (miRNAs), a large class of short noncoding RNAs found in many plants and animals, often act to post-transcriptionally inhibit gene expression. We report the generation of deletion mutations in 87 miRNA genes in Caenorhabditis elegans, expanding the number of mutated miRNA genes to 95, or 83% of known C. elegans miRNAs. We find that the majority of miRNAs are not essential for the viability or development of C. elegans, and mutations in most miRNA genes do not result in grossly abnormal phenotypes. These observations are consistent with the hypothesis that there is significant functional redundancy among miRNAs or among gene pathways regulated by miRNAs. This study represents the first comprehensive genetic analysis of miRNA function in any organism and provides a unique, permanent resource for the systematic study of miRNAs
